転写因子病下垂体ホルモン複合欠損症の病因に関する研究

研究概要	下垂体特異的転写因子であるPit-1は、成長ホルモン(GH)、プロラクチン(PRL)、甲状腺刺激ホルモン(TSH)遺伝子の発現を促進し、逆にPit-1変異はGH、PRL、TSHの下垂体ホルモン複合欠損症を発症させることが報告されている。この疾患の発症機構の解析は、Pit-1が関与する下垂体特異的遺伝子の発現促進機構の解明につながるものであると考え、私どもは、国内外で報告されている種々の変異Pit-1の標的遺伝子活性化能について検討した。 Pit-1の24位のプロリンがロイシンに変化した異常Pit-1の機能を解析したところ、1)CBPへの結合性が欠如していること、2)cAMPによるGH遺伝子発現を促進しないこと認めた。この成績は、CBPあるいはCBPと関連するCBP以外の因子とPit-1は複合体を形成し、この複合体はcAMPによるGH遺伝子発現増強効果に関与している可能性を示唆するものである。一方、Pit-1の271位のアルギニンがトリプトファンに変化した異常Pit-1(R271W)は、最もよく報告されている変異Pit-1であるが、この機能を解析したところ何ら正常Pit-1と変わりはなかった。このPit-1変異例の下垂体ホルモン複合欠損症家系解析からは、この変異が責任遺伝子であることに合致しないケースもあり、この変異の下垂体ホルモン複合欠損症発症における意義については再検討する必要があると考えられた。
