インターロイキン-21(IL-21)による癌の免疫遺伝子治療

研究概要	IL-21は活性化T細胞から分泌され、IL-15と協調的に働きナチュラルキラー(NK)細胞を活性化する。IL-21はさらに、T細胞、B細胞、NK細胞の増殖に関与しており、これら免疫細胞の分化にかかわる因子としても、その機能は重要と考えられる。したがって、悪性腫瘍に対する免疫治療に、IL-21は新しい有効な手段を提供する可能性がある. 本研究では、IL-21の生体内での生理機能を検証する目的で、IL-21遺伝子をマウスにin vivo導入し、NK細胞の活性、生体内における免疫細胞のポピュラリティーと分布、サイトカイン群の産生パターンなどのパラメーターを計測した。さらに、IL-21の抗腫瘍免疫誘導能を解析するために、担癌モデルマウスを作製し、予防、および治療効果を検討した。 IL-21発現ベクター、pGEG.mIL-21とpG.mIL-21を構築した。また同様に、IL-15発現ベクターも作成した。これらをマウスに高圧化に静脈内投与した。このマウスに、悪性リンパ腫細胞、RLmail1を皮下に移植した。IL-21+IL-15投与マウスにおいては正常対照マウスに比し著明な抗腫瘍効果が得られた。脾リンパ球のNK活性を測定すると、IL-15投与により有意に増強していたが、IL-21+IL-15投与によりさらに著明に亢進した。また両サイトカインの共導入により、肝NK細胞の有意な増殖が認められた。担癌マウスの寿命は、IL-21+IL-15治療により有意に延長した。一方、RLmail1をあらかじめ皮下移植することにより腫瘍を樹立させておいたマウスに、後からIL-21+IL-15遺伝子を投与した場合でも、明らかな抗腫瘍効果が得られた。これらの実験結果は、IL-21遺伝子が、IL-15遺伝子と相乗的に作用してNK活性を増強し、悪性腫瘍にたいして予防的、および治療的に抑制したことを示している。
