インターロイキン-21(IL-21)による癌の免疫遺伝子治療

研究概要	IL-21は活性化T細胞から分泌され、IL-15と協調的に働きNK細胞を活性化する。IL-21はさらに、T細胞、B細胞、NK細胞の増殖、分化を促進する。したがって、悪性腫瘍に対する免疫治療に、IL-21は新しい有効な手段を提供する可能性がある。本研究では、IL-21の生体内での生理機能を検証する目的で、IL-21遺伝子をマウスにin vivo導入する実験を試みた。さらに、IL-21の抗腫瘍免疫誘導能を解析するために、肝転移モデルマウスを作製し、予防、および治療効果を検討した。 IL-21および/またはIL-15発現ベクターをマウスに高圧下に静脈内投与したのち、脾リンパ球のNK活性を測定すると、IL-15投与により有意に増強していたが、IL-21+IL-15投与によりさらに著明に亢進した。また肝NK細胞数の有意な増加が認められた。RLmale1悪性リンパ腫を移植し、特異的なCTL活性を測定すると、NK活性とは対照的に、IL-21遺伝子単独投与でもCTLは顕著に増強し、IL-15はそれに対して相乗的に働くことはなかった。サイトカイン遺伝子共導入をおこなったのち腫瘍細胞を経血管的に移植すると、転移巣の著明な減少が得られた。一方、RLmale1を移植することにより転移性肝癌をあらかじめ樹立させたマウスに、後からIL-21+IL-15遺伝子を投与した場合でも、著明な腫瘍抑制効果が認められた。マウスの生存期間は、両サイトカイン遺伝子の共導入群でのみ有意に延長し、予防的投与では80%、治療的投与でも40%のマウスが、完全に腫瘍を拒絶し腫瘍死を免れた。これらの実験結果は、IL-21遺伝子とIL-15遺伝子が相乗的に作用して抗腫瘍細胞性免疫を増強し、悪性腫瘍を抑制したこと、これらサイトカインの投与、または遺伝子導入が、悪性腫瘍の新しい免疫治療に繋がる可能性を示している。
