遺伝子改変モデル動物を用いた雌性生殖管の受精時における分子制御機構の解析

研究概要	受精の分子機構は特にこの20年間に多角的に解析されてきたが、この受精・初期胚発生に関与すると推定される分子群は生体内では多元的に相互作用をしていることが、近年次第に明らかになってきた。このことは、これまでの主に体外受精系を用いた解析では、in vivoにおける受精機構の掌握し様々な不妊症における真の病態生理の解明を目指すには、大きな限界があることを意味している。申請者らは生体内受精環境としての雌性生殖管の生理的機能についての分子生物学的研究を行ってきた。それらの一連の研究の過程で近年、マウスの雌性生殖管に限局的に発現させる遺伝子mogpを解析し、そのcis-regulatory elementを含むプロモーターのDNA断片mogp-2.2を単離した。この遺伝子プロモーター断片を用いると雌性生殖管に限局した遺伝子発現をエストロゲン依存性に誘導することが可能であることをこれまで明らかにした。更に、受精・初期発生における雌性生殖管内の機能分子の働きをin vivoで検討することを最終目的として、その前段階として、まずmogp-2.2を利用して、このDNA断片の下流にCre recombinase遺伝子を連結したベクターを構築し、雌性生殖管内組織にCre recombinaseを限定発現するトランスジェニックマウスの作製を試みている。現在、作製したトランスジェニックマウスの導入遺伝子の発現状態を多角的に解析中である。
