遺伝子改変モデル動物を用いた雌性生殖管の受精時における分子制御機構の解析

研究概要	受精の分子機構は、特にこの20年間に多角的に解析されてきた。受精・初期胚発生に関与すると推定される分子群は、生体内では多元的に相互作用をしていることが近年次第に明らかになってきている。このことは、これまでの主に体外受精系を用いた解析ではin vivoにおける受精機構の掌握し、様々な不妊症における真の病態生理の解明を目指すには、大きな限界があることを意味している。本研究では生体内受精環境としての雌性生殖管の生理的機能に着目し、これまでの生殖生物学的研究を基盤として、卵管特異糖タンパク質遺伝子mogp-1のcis-regulatory elementを含むと推定されるプロモーター領域のDNA断片mogp-2.2等を利用し、遺伝子改変マウスを作成した。作成したmogp-2.2-SV40 T antigenトランスジェニックマウス及びmogp-1欠損マウスの腫瘍形成誘導能、生殖生理学的特性等を多角的に解析したところ以下のことが明らかとなった。1)mogp-2.2はin vivoにおいて雌性生殖管に限局した遺伝子発現能を誘導可能であった。2)その遺伝子発現能はエストロゲン依存性であった。3)mogp-1欠損マウスはin vitroにおいては受精能に低下傾向を認めた。4)しかし、in vivoにおいては、妊孕性の明らかな低下は認められなかった。これらの研究成果は、in vitroとin vivoの受精環境の相違を明確に示しているとともに、複雑な雌性生殖管内における分子制御機構を理解するための基礎データとして意義ある研究成果である。
