アデノウイルスベクターを用いたIGF-1導入による舌筋再生治療の基礎的研究

研究概要	Insulin-like growth factor-1(IGF-1)は、骨格筋の発達、維持、再生段階において重要な役割を持つことが知られている。舌筋は骨格筋と同様横紋筋からなる。外傷や腫瘍摘出により欠損した舌は、各種有茎、遊離皮弁で再建されるが、構語、発語、摂食などに回復はまだまだ十分とはいえず、その治療方法が望まれている。我々は、IGF-1遺伝子を組み込んだアデノウイルスベクターを、舌筋半切モデルラットに投与する事により、IGF-1の舌筋への再生効果について調べる目的で本実験を計画した。ラットIGF-1遺伝子を組み込んだアデノウイルスベクターを本実験では使用した。L6細胞(neonatal rat myoblast cell line)において、IGF-1アデノウイルスベクター感染効果について培養レベルで検討した。またラット舌筋の切除モデル(ラット舌前1/2を切除)を作製し、切除舌筋の組織変化および筋再生について検索をおこなった。さらに再生骨格筋のマーカーであるanti-BF-45(anti-embryonic myosin heavy chain monoclonal antibody)抗体を用い、免疫染色により再生骨格筋の定量をおこなった。その結果、L6細胞への感染実験では、IGF-1濃度は感染ウイルス濃度(MOI)に依存して上昇し、骨格筋線維のhypertrophy,およびdifferentiationを誘導することが示された。ラット舌切除モデルにおいても、BF-45陽性骨格筋線維の増加が認められ、骨格筋再生が認められた。我々はin vitro、およびin vivoにおいて骨格筋に対するIGF-1の遺伝子導入に成功した。アデノウイルスベクターは発現効率が高く、高いタイターを得ることが可能であり、骨格筋細胞のような非分裂細胞に一過性に遺伝子を発現させたい場合に有用なベクターと考えられる。
