マクロファージの新規タンパク因子による骨内グルタミン酸シグナリング調節機構

研究概要	マクロファージ/ミクログリアの産生分泌するNMDA受容体を介したシグナリング増強因子の作用メカニズムを明らかにする目的で、前年度(平成14年度)の結果を踏まえて以下の実験を行った。 (1)幼若ラットの大脳皮質より急性単離したニューロンを用いてホールセルモードのパッチクランプを行い、Yチューブ法を用いて適用したNMDAによって惹起される内向き電流に対するマクロファージ/ミクログリアの培養上清(MCM)の影響を解析した。様々な膜電位におけるNMDA電流のピーク値を計測し、電流-電圧曲線を作成した結果、MCMは測定した全ての膜電位においてNMDAによって惹起される内向き電流を有意に増大することが明かとなった。さらに、Yチューブ法を用いて適用したグルタミン酸によって惹起される内向き電流に対するMCMの影響を解析し、NMDAによって惹起される内向き電流に対する影響との比較検討を行った。その結果、MCMはグルタミン酸電流をNMDA電流よりもより強く増大し、その差は有意であることが明かとなった。 (2)仔ウサギの四肢長管骨より調製した破骨細胞において同様にパッチクランプを試みたが、安定したギガーシルが得られる効率が悪く、Yチューブ法を用いて適用したグルタミン酸によって惹起される内向き電流に対するMCMの明らかな影響を観察するに至らなかった。今後、破骨細胞を単離するのに用いるプラスチックシャーレをガラスシャーレに変えるなど、破骨細胞のシャーレへの接着を弱くする工夫が必要と考えられる。以上の結果より、中枢ニューロンにおけるMCMのNMDA受容体を介したシグナリング増強メカニズムはMgによるNMDA受容体チャネル阻害の緩和作用ではなく、NMDA受容体に対するグルタミン酸の親和性の増大に基づくことが強く示唆された。一方、破骨細胞におけるグルタミン酸シグナリングに対するMCMの作用は今回の実験期間内では明らかにできなかったが、MCMは中枢ニューロンの場合と同様に破骨細胞におけるグルタミン酸シグナリングを修飾する可能性は高いと考えられる。
