OK-432の活性成分を用いた局所樹状細胞療法の開発;Toll様受容体4の関与

研究概要

リポタイコ酸関連分子OK-PSAは、A群溶血性レンサ球菌由来抗腫瘍免疫療法剤OK-432より分離した活性成分であり宿主免疫能の増強を介して抗腫瘍効果を発現する。未成熟な樹状細胞(DC)は癌細胞を取り込み、成熟するとT細胞に癌抗原を提示し抗癌作用を発現する。癌治療においてDCを用いた免疫療法が注目されているが、DCが抗腫瘍効果を発現するためにはDC成熟化の誘導が重要である。本研究ではOK-PSA併用DC局所投与療法開発を目的とし、初年度は、OK-PSAがDCの成熟化を誘導するか否かをi vitro実験系にて検索した。OK-PSA刺激により健常人の末梢血由来DCにおいて、主組織適合性複合系抗原(MHC)classII、CD80、CD83およびCD86の発現上昇およびサイトカイン、ケモカイン産生が認められた。OK-PSA刺激DCは、アロT細胞増殖を刺激し、抗原特異的細胞障害性T細胞(CTL)ならびにinterferon(IFN)-γ産生T細胞を誘導した。OK-PSAによる上記表面抗原の発現増強およびサイトカイン産生は抗Toll様受容体(TLR)4抗体により、ほぼ完全に中和された。TLR4は補助受容体MD-2と複合体を形成し細菌成分を認識する。口腔癌患者由来DCにおいて、TLR4およびMD-2の両遺伝子を強発現している患者においては、OK-PSAは成熟DCを誘導し、この成熟DCはCTLを誘導したが、TLR4あるいはMD-2低発現患者由来DCはOK-PSAで刺激してもCTLを誘導しなかった。OK-PSAが健常人および担癌患者においてTLR4/MD-2複合体を介してDCを成熟化する事が明らかとなった。来年度は担癌マウスを用いた実験にてOK-PSA併用DC局所投与療法の抗腫瘍効果につき検討する。さらにTLR4欠損マウスを用いて同療法の効果におけるTLR4の役割についても検討する。
