光学活性な二核鉄錯体による部位特異的な二本鎖DNA切断と人工制限酵素への応用

研究概要

本研究は標的生体高分子に二本鎖DNAを選び,光学活性なジアミンから誘導される二核鉄錯体による二本鎖DNAの目的位置を選択的に切断する人工制限酵素の開発を行い染色体DNAの機能制御を目指す.
1,8-anthracenedialdhydeとN',N'-dipyridylmethyl-R,R-cyclohexane-1,2-diamineとから誘導される光学活性な複核配位子1,8-(N-anthranylmethyl)-bis(N',N'-dipyridylmethyl-R,R-cyclohexane-1,2-diamine)(1)を合成した。構造の帰属はNMR、質量分析により決定した。
pUC19プラスミドDNA0.57μgを用い、50mMトリス塩酸緩衝液中アスコルビン酸存在下1及び硫酸第一鉄(モル比1:2)を混合し(以後鉄錯体1と呼ぶ)37℃で1時間インキュベートした。その後エチジウムブロマイド含有1%アガロースゲルを用いて電気泳動を行い、蛍光強度比からDNA切断活性能を評価した。実験は全て空気中で行った。
1)鉄錯体1の濃度が5μMでは完全にForm Iの消失が観測された。
2)次にインキュベート時間とDNA切断反応の関係を調べたところ、鉄錯体1はインキュベート直後からForm IIIが生成した。反応開始1分後form Iが消失し30分後では完全にForm IIIだけが観測された。
次にpUC19DNAを用いPCR法により調製した253塩基対フラグメントを用いてDNA切断を行った。鉄錯体1はAT-richな部位を切断していることがわかった。
これらの結果から、鉄錯体1はDNAの二本鎖を同時に、かつ、塩基特異的に切断している可能性が高いと考えられる。
