薬物設計のための疎水相互作用場の構築

研究概要

化合物の水とn-オクタノールの間の分配係数(P)は、医薬品、農薬の分子設計や環境汚染物質の疎水性を定量的に表現する尺度として広く用いられている。
本研究ではこれまでの成果に基づき、疎水相互作用の定量的尺度であるlog Pについて、理論計算および高精度実測法を併用することによってその要因を原子・電子レベルから明らかにし、その結果に基づき薬物分子周辺の疎水相互作用場を定義し、定量的構造活性相関解析法に組み入れ、創薬等への応用をはかることを目的として、以下の成果を得た。
(1)幾つかの有機分子のlog P値(水と各種有機溶媒)について,非経験的分子軌道法-連続体溶媒和理論を適用し,分配のエンタルピー項については溶質-溶媒分子間の静電エネルギー差等,エントロピー項については溶質分子の接触表面積と良好な線型関係が存在することを見出し,分配過程において,溶質分子の水素結合能がその過程に寄与していることを明らかにした。
(2)創薬への応用として、log P値から誘導される記述子を開発し、HIV proteaseの阻害剤のvirtual screeningにおいてその有効性を定量的に示した。この結果は、薬物分子とその標的受容体との結合において、疎水性分布の相補性が要求されることを確認するものである。さらにlog Pの応用研究として、母乳と胎児の血漿中の薬物濃度比について、log P値を用いた解析を行い、log PがM/P比と定量的相関があることを見出した。この結果は臨床結果へlog Pの適用可能性を示唆するものである。
(3)フロー法(流量比法)による分配係数測定の装置および解析法を確立し、従来のフラスコ振とう法では測定が困難であったフラン誘導体、(1)と関係して水素結合能で興味がある含窒素芳香環化合物の分配係数の測定を行い、今後のlog P-薬物分子の疎水性解析に有用な結果を得た。
