新規コレステロール結合プローブを用いた細胞内ラフトの可視化システムの構築

研究概要	我々は、これまでに、コレステロールに特異的に結合する毒素(θ毒素)を改変、無毒化することによってコレステロールに特異的なプローブ(BCθ)を作製し、BCθが脂質ミクロドメイン(ラフト)中のコレステロール密度の高い領域に選択的に結合することを見いだしている。さらに、BCθが細胞表面ラフトの可視化に有用であることを明らかにしている。そこで、本研究では、細胞表面及び細胞内ラフトの同時可視化を目指して、細胞内ラフトの可視化系の構築を進めた。昨年度の研究で、毒素のコレステロール結合ドメインであるC末端のドメイン(D4)と緑色蛍光蛋白質(Enhanced green fluorescent protein ; EGFP)との融合蛋白質(EGFP-D4)遺伝子を作製して大腸菌内でEGFP-D4蛋白質を生合成させ、これを精製してその性質を調べた。その結果、EGFP-D4はコレステロールに対する特異的結合能およびラフトへの選択的結合能を保持していることを明らかにしている。そこで、次に、動物細胞内へのEGFP-D4遺伝子の導入を行った。テトラサイクリンの除去で目的遺伝子の発現が誘導されるように設計されたベクターにEGFP-D4遺伝子を組み込み、マウス線維芽細胞に導入した。薬剤耐性を指標に遺伝子導入細胞を選択的に増殖させ、EGFP-D4蛋白質の発現量の異なる幾つかのクローンを得た。細胞内でのプローブの局在を調べたところ、一部の細胞内小胞に強い染色が認められ、生化学的な解析の結果とあわせて細胞内ラフトへの結合が示唆された。
