LAMP-2によるオートファジーおよび受容体輸送の制御機構

研究概要	我々は、リソソーム膜蛋白質の1つであるLAMP-2の機能解明を目的とし、LAMP-2欠損マウスを作成し、LAMP-2欠損が蛋白質分解能低下とそれに伴う未成熟なオートファゴソームの蓄積、心機能低下を引き起こすことを明らかにした。一方、リソソーム膜にはLAMP-2と構造上極めて類似した蛋白質としてLAMP-1が発現している。我々は以前、LAMP-1欠損マウスを作成し表現型を解析したところ、種々の臓器におけるLAMP-2の発現量の増加以外に正常マウスと比べ際立った表現型は観察されなかった。この結果は、LAMP-1欠損をLAMP-2が機能相補出来ることを示唆している。本研究は、LAMP-1およびLAMP-2のダブルノックアウトマウス(LAMP-1^<-/->/LAMP-2^<-/->)を作成しそれぞれのシングルノックアウトマウスとの比較解析を通して、LAMP-2欠損により引き起こされるオートファゴソーム蓄積の分子機構ならびにLAMP-2のより詳細な生理機能の解明を目的とした。その結果、LAMP-1およびLAMP-2シングルノックアウトマウスと異なり、LAMP-1^<-/->/LAMP-2^<-/->は胎生致死であった。またLAMP-1^<-/->/LAMP-2^<-/->胎児繊維芽細胞の細胞内にはオートファゴソームの蓄積が観察されたが、LAMP-2ノックアウトマウスで観察されたような未成熟なオートファゴソームではなく成熟型であった。この結果からも予測できるように、LAMP-1^<-/->/LAMP-2^<-/->胎児繊維芽細胞における蛋白分解能は正常であった。興味深いことに、LAMP-1^<-/->/LAMP-2^<-/->胎児繊維芽細胞では細胞内にコレステロールの蓄積と脂肪滴形成異常が観察された。これらの結果より、LAMP-2は脂質代謝と或は輸送を調節することにより、リソノーム形成・維持および機能発現に寄与していることが示唆された。
