研究概要	筋萎縮性側索理化症(ALS)の前角細胞のリボゾームRNA遺伝子転写活性の減少メカニズムを解明する目的から、下記の検討を行った。 1.ALS脊髄前角細胞における粗面小胞体の数 ALS前角細胞の、大きさ1ミクロン以上の粗面小胞体を計測し、クロマトリーシスを示す細胞のみならず、萎縮して濃染する細胞でもその数が減少していることを見出し報告した。 2.ALS前角細胞における粗面小胞体の超微構造の観察 ALS前角細胞の粗面小胞体は、変性初期の、クロマトリーシスを示す細胞では断片化し、変性末期の萎縮した細胞ではシステルナの不規則な膨化と、小胞体膜状のリボゾームが減少していること(ribosomal detachment)を見い出し報告した。 3.蛋白質の折り畳みに関連する小胞体マーカーKDELから見たALS小胞体の変化小胞体マーカーを用いた免疫染色所見では、一見正常に見えるALSの前角細胞でも、小胞体は袋状の拡大を示し、変性の初期変化と言われているクロマトリーシスを示す前角細胞では細胞辺縁部に細かな断片状に存在し、一方、変性末期と考えられる萎縮した前角細胞では、核の周囲に断片化して存在することを見い出した。
