微小管機能の障害によるアポトーシスへのカルシニューリンの関与についての研究

研究概要	抗癌剤であるタキソールは微小管重合作用を有し、細胞分裂を抑制することでアポトーシスを誘導することが知られている。この誘導作用には、Bcl-2の燐酸化反応が関与していることが報告されていた。今回、培養ヒト膀胱癌細胞を用いてBcl-2の脱燐酸化反応へのカルシニューリンの関与について検討した。実験には悪性度の高いT24細胞と悪性度の低いKK47細胞の二種類の癌細胞を使用した。1.WST-1法による細胞死の検討では、T24細胞はタキソールに対して抵抗性を示した。カルシニューリン阻害剤であるサイクロスポリンA(CsA)とFK506の添加により抵抗性が著明に消失した。KK47細胞ではCsAとFK506の添加効果は弱くしか認められなかった。2.両細胞にカルシニューリンが存在し、その活性は、CsAやFK506の添加により完全に消失した。3.ヘキスト染色とFACScanを用いたサブG1測定法およびカスパーゼ活性測定法により、タキソールによる両細胞のアポトーシスが観察された。さらに、アポトーシスはカルシニューリン阻害剤の添加により増強された。4.Bcl-2の燐酸化反応については燐酸化特異抗体を用いた免疫ブロット法により検討した。タキソール処理によるBcl-2の燐酸化反応は、CsAやFK506の添加により増強された。これらの結果は、カルシニューリンがBcl-2の脱燐酸化反応に関与し、タキソールによるアポトーシスを抑制することを示唆している。また、最近、カルシウム/カルモデュリン依存性蛋白質燐酸化酵素IIが、タウのチューブリン結合部位を燐酸化して、チューブリンとの結合を阻害することで微小管の形成を阻害することを見い出した。また、本酵素で燐酸化されたタウに対する特異抗体を作製した。作製した抗体を用いて、神経細胞のアポトーシスへのタウの燐酸化の関与を明らかにする予定である。
