脳内移動型および非移動型GnRHニューロンにおける新規遺伝子の単離

研究概要

我々はこれまでに、免疫染色によって同定した性腺刺激ホルモン放出ホルモン(GnRH)ニューロン単離技術の開発に取り組み、その技術を確立した。成熟した硬骨魚(シクリッド科)の脳を摘出し、4%パラホルムアルデヒド中で固定後、5%のゼラチン中に包埋し、マイクロスライサーを用いて500μmの厚さの矢状断切片を作成した。脳切片を3種類の抗GnRH抗体と12時間反応させた後、赤色の蛍光を呈するCy3で標識された抗ウサギIgG抗体で免疫染色した。脳切片を蛍光顕微鏡下で観察し、免疫染色されたGnRHニユーロンをマイクロマニピュレーターに接続したマイクロキャピラリーピペットで採取・単離した。単離されたニューロンをプロテアーゼ消化し、抽出された全RNAから単一ニューロン由来のcDNAライブラリーを作成した。
免疫染色および単離されたGnRHニューロン中のGnRH mRNAの状態を確認するため、3種類のGnRH遺伝子に特異的なプライマーを作成し、PCRにて増幅した。PCR増幅されたアンプリコンの塩基配列を確認するため、PCR産物をpGEM-T easyベクターに挿入してサブクローン化し、Genetic Analyzer(ABI)を用いてシークエンスを行った。
その結果、固定・免疫染色後においても単一ニューロン中のGnRHm RNAは保存されており、生理学的な解析を行うために十分量なmRNAが得られることが示唆された。
