ラット海馬ニューロンの長期増強(LTP)に対する実験的脳外傷の影響の解析

研究概要

外傷後のてんかんや記憶障害について実験的脳外傷モデルラットを用いて調べた。麻酔下に、ウィスター系の成熟雄ラット(250〜300g)の左側の頭蓋に約3mmの穴を開窓し、脳に直接、圧衝撃(Fluid percussion injury : FPI)を与えた。その後1〜2週間通常の飼育を行い、断頭し、衝撃直下の冠状断海馬スライス標本および、:Bregna-5〜-6mmの水平断海馬スライス標本を作製した。
結果:1)電位感受性色素を用いた光学的電位記録法により歯状回、海馬CA3-CA1における活動電位の伝播を記録した結果、障害された大脳皮質直下の海馬では外傷側のニューロン活動性とその伝播領域が、非外傷側に比べ著明に抑制されていた。2)一方、外傷部位から離れている脳底部付近の(側頭葉皮質に隣接する)海馬では、非外傷側に比べ外傷側の海馬ニューロンの活動性はむしろ強く亢進していた。また、細胞外電極法により得られたfield potentialは、外傷側において反復性のスパイクを持ち、外傷側で明らかに興奮性の増大が認められた。3)細胞内電極法(一部パッチクランプ法)により、興奮性シナプス後電位(EPSP)の時間経過(time course)が著明に延長し、この延長したEPSPにようて活動電位が繰り返し惹起された。3)海馬ニューロンの興奮性増大は、GABA_A受容体阻害剤であるビククリンを投与すると、さらに著明に増強した。このEPSPの延長と反復性活動電位の発生は正常なラットと同様な程度で観察されたことから、FPIラットにおける興奮性の増大は抑制神経の障害による脱抑制によってもたらされたものではなく、興奮性神経のシナプス伝達が直接、過剰興奮を起こしているものと考えられる。今後、グルタメートの過剰放出などによる神経障害や長期増強の障害について調べ、てんかんや記憶障害の機構について検討する。
