ラット海馬ニューロンの長期増強(LTP)に対する実験的脳外傷の影響の解析

研究概要

中等度の液体圧衝撃をラットの左頭頂部大脳皮質に与えて作成した実験的脳損傷モデルラットfluid percussion injury(FPI)ratから、海馬領域を含む厚さ約400ミクロンの水平断脳スライス標本を作製した。この実験的脳損傷モデルラットの海馬ニューロンにおいて、刺激の大きさと反応(EPSPの立ち上がり速度:EPSP slope)の関係(input-output relation)をみたところ、FPIモデルラットの衝撃側で、興奮性シナプス伝達が抑制されていた。しかし、神経細胞の静止および興奮時の電気生理学特性には健常ラットと差異はなく、障害ラット脳の左右差も認められなかった。また、興奮性シナプス後電位(EPSP)は傷害側でtime courseが延長し、頻回の活動電位が発生した。興奮性シナプス伝達の長期増強(long-term potentiation:LTP)と長期抑圧(long-term depression:LTD)は消失することはなかったが、健常ラットに比べ明らかに抑制されていた。FPIモデルラットでのEPSPの抑制は必ずしも器質的障害はないが、衝撃による機能低下が起きていることが推察される。脳外傷や虚血(低酸素症)時にはアデノシンが放出され、グルタミン酸の過剰放出による神経障害を防ぐ効果(脳保護効果)を持つと報告されているが、このような抑制機構が働いているものと思われる。しかし、興奮性シナプス伝達のLTPが明らかに両側ともに抑制され、LTDが比較的多く観察された。アデノシンと類似の脳保護作用を持つことが示唆されているアンソラキノン誘導体(イモデインやレイン)はFPIモデルラットにおいて緩徐であるが可逆性にグルタミシ酸遊離を抑制する作用を持つことが示唆され、これが脳保護作用のメカニズムである可能性が示された。
