T細胞ホメオスターシス増殖の調節機構の解明

研究概要	本年度の研究では、平成14年度の研究成果をさらに発展させて、ヒトT細胞においても活性化に伴うCTLA-4の発現誘導にIL-2が重要なことを見い出した。またin vivoでのCTLA-4発現誘導におけるIL-2の重要性とホメオスターシス増殖の制御における同機構の役割を検討した。MHCクラスI拘束性の単一のT細胞レセプター(TCR)を発現するコントロールTCR-RAG-2KOマウスおよびTCR-RAG-2KO-IL-2KO、TCR-RAG-2KO-IL-15KOマウスからナイーブT細胞を分離して、それぞれのT細胞をRAG-2KO、RAG-2KO-IL-2KO、およびRAG-2KO-IL-15KOマウスに移入した。経時的に末梢血を採取し、検出されるT細胞の数を指標にして、生体内でのホメオスターシス増殖におけるIL-2の重要性を検討した。その結果、TCR-RAG-2KO T細胞をRAG-2KOマウスに移入した系に比べて、同T細胞をRAG-2KO-IL-2KOマウスに移入した場合にはホメオスターシス増殖の有意な亢進が、逆にRAG-2KO-IL-15KOマウスに移入した際にはホメオスターシス増殖の低下が観察された。その他の系では顕著な差異は観られなかった。このことから、ホメオスターシス増殖の促進と抑制には、おそらく樹状細胞由来のIL-2とIL-15がそれぞれ重要であることが示唆された。
