研究概要

本研究では、近年社会問題化している青少年における感情制御の未成熟、中高年におけるうつ病および自殺の急増に対して、その基盤となっている感情制御の脳内メカニズムおよび感情障害の病態を最新の脳科学的研究手法を用いて解明し、その予防法、治療法の確立を目的として、わが国の代表的な研究者を結集して、以下のような企画調査研究を行い、特定領域研究の研究領域として発展させるべく検討を行った。
1)脳機能画像解析装置(PET, MRI, MEG)を用いた研究
本研究におけるfMRIを用いた検討で、不快刺激の予測に右前頭前野が、快刺激の予測に左前頭前野が関与することが明らかとなった。うつ病では左前頭前野の機能低下があることから、快の予測が困難となり悲観的思考になると推測された。
2)感情障害患者のリンパ球および死後脳を用いた研究
大うつ病、双極性障害の患者血球細胞を用いた研究で、細胞内カルシウム動態の異常とミトコンドリア遺伝子多型の関連性が示唆された。また、うつ病の死後を用いた研究ではG蛋白機能や転写調節因子の異常性が確認された。
3)動物モデルを用いた研究
恐怖条件付モデルラットを用い、不安行動と脳内ドパミン、セロトニン神経活動の関連性を検討した。また、新生児期母子分離させたラットに成熟後拘束ストレスを負荷すると正常飼育群に比較して、行動量低下、コルチコステロン高値が認められ、ストレス脆弱モデルとして妥当であることが確認された。このストレス脆弱モデルラットにおいて、cDNA Arrayを用いて遺伝子検索をした結果、c-Jun N-terminal Kinase 2(JNK2)の発現が低下しており、新生児期の母子分離ストレスが遺伝子を介して成熟後のストレス脆弱性を形成する可能性が示唆された。この結果は、PTSDなどのストレス性精神障害のリスク要因として幼少時期の心的外傷(両親の離婚、虐待)の存在が報告されていることと一致し、この時期の母子関係の重要性を遺伝子レベルで解明できる可能性を示唆している。
以上のような研究成果を踏まえて、特定領域として研究を発展させたい。
