原子間力顕微鏡を用いた光合成アンテナ色素蛋白複合体超分子配置のイメージング

研究概要	光合成細菌の光合成系は二種類のアンテナ色素蛋白複合体(LH1及びLH2)と光反応中心(RC)の3つの色素蛋白複合体が有機的に結合した超分子配置を形成することにより生理機能を発現している。光合成膜蛋白質の結晶構造解析の成功により、RC, H1,LH1-RCコア複合体,LH2の構造が原子スケールで明らかにされつつある。LH1,LH2の2種類のアンテナ複合体の空間配列については、物性データを解釈する際のモデルとして便宜的な構造が仮定されている。しかしながら、「実際に生理機能を有する(生きている)光合成膜でアンテナ複合体の空間配置はどのようになっているのか?」と言う素朴な疑問については、光合成初期過程の機構解明の最も基本的な概念であるにも関わらず、現在のところその回答を見出そうする機運すら存在しない。本研究の目的は、原子間力顕微鏡(AFM)を用いて生理活性を有する光合成膜のその場観察を行い、集光性色素蛋白複合体の超分子配置のマッピング行うことである。今年度は、その手初めとしてAFM測定に最適と考えられる平坦な光合成膜を持つ紅色光合成細菌Rps. viridis及びRps. acidophilaから調製した光合成膜試料に焦点を絞り、画像計測の物差しを作成することを目的として研究を行った。Rps. viridisにはアンテナ複合体としてLH1複合体の一つしか存在しないことが知られている。AFM計測を行ったところ、初期的ではあるがこのLH1に対応すると考えられる直径約100Å程度のリング画像の取得に成功した。Rps. acidophilaではこの大きなリングの他にLH2に由来すると考えれるサイズの小さなリング画像が混在している初期イメージが取得できた。独立な手法を用いて得られたAFM画像の同定を行う目的で、透過型電子顕微鏡を用いた光合成膜の観察を開始した。紅色光合成細菌Rps. pulstrisとRb. sphaerodiesから調製した光合成膜に関して初期的データの取得に成功しており、現在、シミュレーション等を行うことにより得られた画像が色素蛋白複合体のものと考えて妥当かどうかの検討を行っている。
