研究概要

概日リズムは多くの生物に見られる行動やホルモン分泌などをはじめとした生体リズムである。このリズム形成には多くの分子が関わっている。本研究では昆虫で行動リズム伝達を制御する、18アミノ酸からなる神経ペプチド、Pigment-dispersing factor (PDF)の細胞核内レセプター(受容体)の同定を夜行性昆虫フタホシコオロギで試み、PDFの核内での相互作用の相手として有力視されているTIMELESSタンパク質のcDNAクローニングと発現を中心に以下の実験を実施した。
1:昆虫TIMELESSのcDNAクローニング
PDFの相互作用の相手として有力であるTIMELESSタンパク質(全長約1200 AA)のcDNAクローニングを夜行性昆虫フタホシコオロギと、同じく夜行性昆虫であるカイコガで行った。その結果、フタホシコオロギでは約400 AAをコードする部分構造が明らかとなった。一方、カイコガでは全長配列(ca.1200 AA, ca.4200 bp)の解析に成功した。現在、フタホシコオロギの全長配列の解析と並行し、カイコガTIMELESSのS2細胞による発現を実施中である。
2:フタホシコオロギpdfのゲノム解析
フタホシコオロギPDF前駆体は他の昆虫PDF前駆体と比較するときわめて特異な構造をしている。それはN末端に分泌シグナルを持たないこと、前駆体構造が異常に短いことである。特に分泌シグナルではなく、塩基性アミノ酸に富んだ核移行シグナルをもつ。今回、この特異な構造をより詳細に解析するためにフタホシコオロギpdfのゲノム解析を行った。その結果、cDNAクローニングにより得られた同一の配列が得られた。しかしながら、5'非翻訳領域にゲノム配列とmRNA配列に相違が見られ、これがスプライシングサイトであることが判明した。つまりフタホシコオロギpdfは複数のエキソンにまたがってコードされていることが明らかとなった。このことは、現在までにゲノム解析が実施されているショウジョウバエやガンビエハマダラカでは見られず、昆虫では初めての例である。
