カシノナガキクイムシの穿孔を阻害する天然化合物の探索

研究概要

14年度の研究において、カシノナガキクイムシ(以下、カシナガ)が穿孔することによって辺材部に形成される壊死変色部では、ガロ酸とエラグ酸の含有率が増加していることが明らかにされた。
辺材部にこれらタンニン酸の水溶液をしみ込ませたバイオアッセイの結果、ガロ酸単独では辺材部の含有率(材湿重%、以下同じ)の約40倍にならないと、穿孔を完全に阻害することができなかった。それに対して、エラグ酸の場合、辺材部の半分の含有率で完全に穿孔を阻止することができた。これらの結果は、前年の穿孔によって形成された壊死変色部をカシナガ成虫が避けるのは、エラグ酸が至近要因であることを示唆している。しかし、実用化試験を考えた場合、エラグ酸は水に対する可溶性が低いという難点が残された。また、樹皮に塗布した場合には、忌避効果は認められなかった。樹皮に塗布した場合、タンニン酸が結晶として析出してしまうことが、穿孔を阻害することのできない原因と考え、現在も溶剤等を検討している。
エラグ酸、ガリック酸を異なる濃度に調整して添加した培地上において、病原菌Raffaelea quercivora(8菌株)、カシノナガキクイムシの菌のうから分離された酵母(8菌株)の菌糸伸長量を比較した。その結果、いずれの菌株も、有意な成長阻害は認められなかった。これらの結果は、カシナガがこれらのタンニン酸を忌避する究極要因は、餌としているアンブロシア菌類が育たないためではないことを示している。
