抗酸化物質を用いた新たな神経突起・神経細胞分化誘導法の開発

研究概要

抗酸化剤が脳機能の改善・防護に関する機構のメカニズムを知るために、様々なニトロンスピントラップ剤のPC12細胞の神経突起形成に対する効果を検討した。実験に用いたスピン捕捉剤はPBN(I), S-PBN(II), POBN(III), DMPO(IV), M4PO(V), DEPMPO(VI), DBNBS(VII)を用いた。(I)〜(III)はフェニル基を持つもの、(IV)〜(VI)は5員環状ニトロンスピン捕捉剤、(VII)はニトロソスピン捕捉剤である。本年度は、その各々の薬剤の分化効率を評価し、どういった構造、すなわち5員環構造か6員環構造を持つ物質か?疎水性構造か親水性構造化か?などの性質が分化に重要か検討した。結果はPBN、POBN、S-PBNの順の神経突起形成の効率であり、PBNが最も効率が高かった。また、DMPO, M4PO, DEPMPO, DBNBSについては全くそう言った効果は示されなかった。この結果は、より疎水性の高いニトロン系スピン捕捉剤が神経突起の形成には重要であり、ドラッグデザインにおいて重要な知見であると考えられる。平成15年度はシグナル伝達の側面から、このメカニズムについて検討を加える予定である。具体的には神経突起形成に必要であると言われているPKCε活性化とRasとその下流のシグナル分子MAPキナーゼファミリーのERK活性化などの各種キナーゼの活性化、機能的な面としてドーパミン生合成への効果も検討する。
