成体の脳室下細胞と脈絡叢上衣細胞より神経幹細胞を樹立する

研究概要	(1)脳室下組織からの幹細胞の樹立これまでの幹細胞の樹立は主に胎児あるいは生後間も無い若い動物の脳からであった。本研究では、死後の成体の脳室下組織から幹細胞の樹立が可能かどうかを調べた。成体及び若年(生後1〜7日)のラットを用いた。動物を過剰な麻酔で殺し、冷蔵庫で1-6日保存した。成体では側脳室外側部(線状体側)の脳室下組織を、若年ラットでは脳組織全体をソースとして用いた。驚くべきことに、死後2日迄のラットからは生体の場合と同様の収率で細胞塊(neurosphere)が得られた。当然のことながら、若年ラットからは成体の場合よりも多くの細胞塊が得られた。成体では死後4日まで、若年では死後6日まで少数ながら細胞塊が得られた。細胞塊からはの細胞分化の様式は、死体から得られたも生体からの場合と全く同じであった。死体から得られた細胞塊を脊髄に移植すると細胞は良く生着した。このように死体からの幹細胞も生体と同様の増殖分化能を持っていることが明かとなった。これは神経幹細胞の臨床的な応用というで大きな意義がある。 (2)脈絡叢上衣細胞からの神経幹細胞の樹立脈絡層は成体ラットと生後7日のラットを用いた。脈絡叢上衣細胞にはBrdUの取り込みが見られるので、成体でも分裂する細胞であることが分る。この組織から神経幹細胞がとれるかどうかを調べた。まず、細胞の標識のために脈絡叢そのものを抗原としてモノクローン抗体を作製した。得られた抗体の中から脈絡叢上衣細胞を特異的に認識する抗体を選んで、蛍光色素を結合させて脈絡叢の上衣細胞を標識した。標識された細胞をセルソーターで分離し、細胞培養系に移して、幹細胞としての性質つまり細胞塊(neurospphere)を作る性質があるかどうかを調べた。少数ではあるが、細胞塊が形成されることが明かとなった。この結果は脈絡叢の上衣細胞の中に幹細胞の性質をもつ細胞があることを示すものである。
