種々の環境発癌因子の標的分子の同定―「分子標的予防法」に向けての基礎的研究

研究概要	ヒトの発癌に関与する因子は、何らかの作用を遺伝子の機能に影響を及ぼしていると考えられ、我々は、特に遺伝子発現への発癌因子の影響に着目した研究を本研究課題で実施してきた。そこで、発癌因子の作用が予測される遺伝子として、発癌抑制機構として中心的な役割を果たしているp53-RB経路に着目し、その経路に関与する発癌抑制作用を有する遺伝子を利用した。我々は、従来から有していた多数の細胞周期抑制遺伝子、アポトーシス誘導遺伝子のプロモーター・スクリーニング系に、p19とSiah-1遺伝子のプロモーターを追加し、環境発癌因子によるこれら遺伝子プロモーターへの作用を検討した。その結果、環境発癌因子であり、かつ発癌プロモーターの12-0-tetradecanoylphorbol-13-acetate(TPA)がp18遺伝子発現を抑制し、細胞増殖を促進することを見い出した(Oncogene, Matsuzaki, Y., Sakai, T. et al.)。近年、p18遺伝子が欠失すると様々な組織で高頻度に腫瘍が生じることが示され、少なくともマウスにおいては、p18が癌抑制遺揖子として機能していることが明らかとなった。さらに、最近の知見では、p18遺伝子の2つのアリルのうち片側が欠失するだけで、発癌物質によって生ずる癌の頻度が著しく増加することが判明した。そして、発生した癌を解析したところ、p18遺伝子は片側のアリルが欠失しただけで、もう一方は正常に保たれていた。この結果は、p18遺伝子の発現が量的に減少するだけで、細胞は癌化へ向かう可能性を示唆する。我々の結果は、TPAが、発癌抑制機構として中心的役割を果たしているp53-RB経路に存在するp18の発現を抑制することで細胞増殖を促進し、癌化を引き起こす可能性を示唆する。
