研究概要

ヒトDNase IIの遺伝的多型はわれわれの研究室で発見された。DNase II多型は、優劣のある、常染色体の2対立遺伝子DNASE2^*HおよびDNASE2^*Lによって支配され、酵素活性の強弱によって3種類の表現型に分類できる、極めて珍しい遺伝形式を示す。その後、われわれは世界に先駆けてDNase II遺伝子クローニングに成功し、遺伝子座位が19p13.2-13.1に位置することを解明した。そのほか、DNase IIのスプライシング機構も明らかにしている。しかし、この遺伝子のエクソン部分やイントロン部分の配列に遺伝的多型の原因を求めることはできなかった。したがって、この遺伝子の5'上流域や3'下流域のヌクレオチド配列を解明することを研究の目的とした。
DNase IIは活性強度に関する遺伝的多型性を有するが、その分子論的基盤として、(1)DNase II一次構造上のアミノ酸置換に基づく比活性の相違、または(2)プロモーター領域における変異による転写活性の変動に基づく酵素産生量の相違が想定された。初めに、DNASE2^*HおよびDNASE2^*Lホモ接合体のDNase II遺伝子解析から、open reading frameに相当するエキソン領域には両者間に相違は認められず、同一の塩基配列を持つことが明らかとなった。従って、(1)の仮説は否定された。次に、DNase II遺伝子構造に関する知見に基づき、その遺伝子上流の塩基配列を-2379残基まで決定した。そこで両対立遺伝子由来の遺伝子上流領域(-2229から79残基まで)のプロモーター活性をtransient transfection luciferase assayによって検討した。その結果、HepG2などの検査したすべての細胞において、DNASE2^*HはDNASE2^*Lに比較して5-30倍プロモーター活性の高いことが判明した。一連のdeletion reporter constructのプロモーター活性を調べたところ、両者の相違は近位プロモーター領域に存在していた。両者の塩基配列の比較から、-75残基において塩基置換が認められ、DNASE2^*HではG残基が、DNASE2^*LではA残基が同定された。
