皮質内微小電極を用いた大脳機能カラム直接刺激による人工視覚再建

研究概要

全盲ラット動物モデルにおいて皮質内電気刺激による変化を行動学的に解析した。行動の定量化にはvideo motion analysisを用いた。円形のオープンフィールドに正常ラットを移し、種々の環境光下でその行動を定量的に解析した。その結果、光の強度に依存して,逡巡行動、探索行動、周回行動それぞれにおいて明るさが250luxで抑制されることが判明した。これらのラットの視神経を経前頭法で両側切断、全盲動物モデルを作成し、両側の一次視覚野と感覚運動野に定位的に皮質内電極を挿入した。電極先端は皮質カラムの第4層にあたるようにした。電極はラット頭蓋に装着したソケットより、種々の条件で電気刺激を加えることができるようにした。このような動物を行動学的に解析したところ、暗環境下の正常動物と同様の行動がみられた。ついで、電極あたり100-500μA、3-60Hzの条件を試みた。500μA、3Hzの感覚運動野刺激では、その行動抑制は認められなかった。しかし、同じ条件で、一次視覚野を刺激すると行動抑制が認められた。これは視覚が得られたのと同様の反応である。さらに、これが視覚特異的なものかどうかを検索するため、光刺激を条件刺激、電気ショックを無条件刺激とした白色ノイズに対するfear potentiationの有無を定量化することにより検討した。これは条件刺激が満たされたときに無条件刺激によって惹起されたfear potentiationが無条件刺激が無いときにも認められるという現象を利用しており、条件刺激に対する特異性に優れた実験行動学的手法である。この結果、皮質内電気刺激が視覚特異的行動変化を引き起こすことが証明された。今後、より複雑な人工視覚反応を形成できるよう電極や刺激方法を発展させたい。
