脳幹からの脊髄内痛覚受容神経回路網への下行性修飾

研究概要

本研究においては、脳幹から脊髄への下行性神経修飾機構、および脊髄レベルにおける痛覚関連ニューロン機能を、神経回路網レベルで検討することを目的とした。そのため、平成14年度には、in vitro脳幹脊髄標本およびin vitro脊髄標本を対象とした高速光計測システムによる膜電位イメージングの方法論を確立した。in vitro脳幹脊髄標本では、標本の生存性について、脳神経(舌下神経根)あるいは脊髄神経(頚髄C4前根)よりの呼吸神経活動モニターを行うことにより確認を行った。平成15年度には、この計測法とin vitro標本とを組み合わせた方法論を応用して、痛覚神経回路網機能に関し、以下の解析を行った。
1.麻酔科臨床において使用されている局所麻酔薬であるリドカインとブピバカインに注目し、それらの脊髄後角内痛覚神経回路網への作用を解析した。この解析により、リドカインとブピバカインは、作用の強さと持続時間に差があるものの、ともに後角内痛覚神経回路網においてシナプス伝達を抑制すること、その作用は浅層(lamina I-II)よりも深層(lamina III-IV)において、より強く認められることを明らかにした。この研究成果は、英文原著論文として発表すべく、現在、原稿を準備中である。
2.最近、神経科学の分野で注目されている細胞外ATPの神経回路網機能への修飾作用に着目し、脊髄後角内の痛覚神経回路網機能に対する細胞外ATPの作用を解析した。細胞外ATPは,後角神経回路網の神経活動に対し、P_2X受容体を介する直接的な増強および分解産物アデノシンを介する抑制の二面的な作用を有すること、細胞外ATPによる神経活動増強作用は浅層部におけるよりも深層部においてより強く発揮されることを認めた。
