複合糖質の新規キラル分析法の開発

研究概要	糖VCDデータベース構築のため、まず単糖を網羅的に測定した。その際、αおよびβアノマーを個別に測定するためにメチルグリコシドをモデル化合物として選択し、それらについても測定した。また、いくつかの二糖や高次のオリゴ糖についても測定した。溶媒はDMSO-d6を用い、特に指紋領域に着目した。その結果、α-グリコシド結合を持つ糖においてのみ、1145cm^<-1>付近において特徴的な下向きの鋭い吸収帯が存在することを見出した。1位の酸素^<16>Oを^<18>Oに置換すると本吸収帯がシフトしたことから、本吸収帯はα-グリコシド結合特異的な吸収帯、すなわち「α-Glycoside band」であることを見出した。本特徴的バンドを用いてα-β比の定量、酵素反応のモニタリングなども検討した。また、DMSO-d6以外の溶媒、ひいては他の波数領域でもVCDを測定した。また、他予算で設置されたVCD装置の調整をおこなった。本装置は、従来装置よりさらに高波数領域の測定が可能である。本装置を使用した測定条件の詳細な検討を行い、糖鎖のC-H、及びO-Hの振動の測定を試みた。糖鎖の結合位置とこの領域のVCD吸収とのある程度の相関が確認できた。一般に糖のVCDは強度が弱くブロードになっており、解析が困難であるように思われたが、1145cm^<-1>付近において糖のα、βを識別する「α-Glycoside band」を見出した。これまでVCDは主に絶対立体化学の決定・タンパクの二次構造解析などに利用されてきたが、本研究はVCDによる糖鎖の解析という新たな研究領域の可能性を示すものである。また、本実験はVCDによる酵素反応モニタリングの初めての例である。
