嗅神経系の髄鞘形成グリア細胞がもつ神経再生促進機能の分子生物学的研究

研究概要	末梢神経に比べ中枢神軽の軸索再生は困難であり、内在的要因や微小環境の違いが問題とされている。しかし嗅覚ニューロンは日常的に死滅と新生を繰り返し、新生ニューロンからは新しい軸索が嗅脳の定位置に伸長しシナプスを形成する。この特殊な軸索再生能は髄鞘を形成するユニークなグリア細胞(olfactory ensheathing glia:OEG)が担っており、損傷脊髄に移植すると著明な損傷修復効果が認められることから、医学的応用が期待されている。本研究はOEGの機能に直結する分子基盤を解明することを目標にしている。昨年度確立したOEGの純粋培養法を駆使して本年度は、OEGの軸索再生能と関連が深い、1)神経栄養因子の産生能、2)細胞運動能とその制御機構を検討し、OEGに特異的に発現する分子を同定するため、3)DNAマイクロアレイ解析、プロテオーム解析を実施した。その結果、1)神経栄養因子と受容体は、NGF、GDNFのmRNA発現は認められたが、BDNF、NT-3のmRNAは検出できなかった。一方、神経栄養因子の受容体についてはp75とTrkc、GFRα1mRNAだけが発現しており、OEGはNT-3とGDNFに選択的に応答することが示唆された。2)OEGはアストロサイトと比較して運動量の大きな細胞であり、運動量はNT-3の作用で減少し、細胞内cAMPを上昇させるforskolin処理に伴って減少した。すなわちOEGの運動量は外的刺激や細胞内の特定シグナル伝達で調節されることが示唆された。3)OEGのDNAマイクロアレイ解析、プロテオーム解析は現在進行中である。OEGに特異的で、軸索再生や細胞運動に関係する分子の同定が期待される。
