RasH2マウスの高発癌性に関わる生理的バックグラウンドの研究

研究概要

rasH2マウスがDMH誘発大腸癌に高感受性である理由は明らかにされていない。前年度はrasH2マウスがDMH誘発大腸癌において導入したc-Ha-ras遺伝子の発現が腫瘍部で著しく亢進していたことから、この遺伝子の発現が高感受性に関与していることを報告した。本年度は腫瘍形成以前にDMH投与に対するマウスの反応性の違いがrasH2マウスの高感受性に結びつくと考え、腫瘍が肉眼的に確認できる以前のDMH投与10週後と16週後にマイクロアレイ(Gene Chip Murine Genome U74A Array v.2)を用いてrasH2マウスと対照のnon-Tgマウスで大腸ならびに肝臓での遺伝子の発現量を比較した。その結果、癌組織が肉眼では検出されない時期の大腸組織において、特にMus musculus mRNA for intelectin, Mus musculus angiogenin-3 precursor mRNAの2種類の遺伝子発現がnon-Tgマウスに、比べてrasH2マウスで上昇しており、Mus musculus angiogenin-3 precursor mRNAの過剰発現が血管新生を促進させることで癌の発生、進行をはやめていること、またMus musculus mRNA for intelectinの過剰発現は、腸内細菌の菌体成分との関係が考えられるが、これについてのさらなる検討が必要であると思われた。また、肝臓ではnon-Tgマウスと比較してrasH2マウスで肝臓での第II相の代謝酵素で、アミン、ステロイド、胆汁酸、発癌物質などの代謝に関与するMus musculus (10-1)hydroxysteroid sulfo-transferaseの発現が特に上昇しており、Mus musculus CYP4A10などのP450(脂肪酸の水酸化により代謝する酵素)の発現が減少した。
