遺伝子・遺伝子キャリアー複合体の細胞内でのトラフィックコントロール

研究概要	細胞内へ外来遺伝子を送達する際、塩基性の遺伝子キャリアーを利用する方法が簡便で一般的な手法となっている。そのキャリアー分子はDNAと複合体を形成し、細胞内へ取り込まれるが、その多くがエンドソームを経た細胞内分解システムへ流れていき、さらに、幸運にも細胞質へ脱出できた複合体も核へ移行できる確率は低く、したがって、細胞内でのエンドソームから細胞質への移行、複合体の解離、そして、核への局在についてそれぞれのステップの効率を向上させることにより、実用レベルに達するような遺伝子キャリアーができると期待される。本研究ではその各ステップを人工的にコントロールし、遺伝子発現効率を向上させる手段を目指すとともに、それを制御することで機能的な遺伝子デリバリーあるいは発現制御を行うテクニックを探ろうというものである。昨年度はエンドソームから細胞質へ移行を促進するペプチドを報告したが、本年度はポリエチレングリコールからペンダント型にカルボキシル基を有する酸性ポリマーを合成し、そのエンドソーム破壊による遺伝子発現効率上昇を実現した。また、この酸性ポリマーを糖修飾することにより、細胞表面レセプター関与での取り込みが認められ、また、ポリエチレングリコールの血中でのステルス性も加わって、in vivoで遺伝子キャリアー分子の構築に成功した。一方、遺伝子発現に必須なDNA複合体の解離について、細胞質中での、解離の可能性について、in vitroの実験を行ったところ、ポリマー性の遺伝子キャリアーの場合は細胞質中のタンパク質成分がその解離に関わっていることが示され、脂質系のキャリアーの場合ではむしろ細胞内の膜成分との相互作用の結果、外来DNAの遊離が達成されていることが指摘された。この結果は脂質系とポリマー系遺伝子デリバリーの違いを理解する上で重要な知見で、今後の遺伝子キャリアー設計に重要な情報となるだろう。
