Naチャネルのシャペロンによる発現調節機構とアンカー蛋白による局在化機構の解明

研究概要

電位依存性Naチャネルの細胞膜発現調節機構におけるシャペロンの役割を解明するために、発生学的に神経堤に由来する培養ウシ副腎髄質クロマフィン細胞を用いて実験を行い、以下の結果を得た。
(1)熱ショック蛋白90(HSP90)をゲルダナマイシンによって阻害すると、Naチャネル発現量は変化しないが、Naチャネルの機能は亢進する。(2)高濃度グルコースにより細胞膜におけるNaチャネル発現量は変化しないが、Naチャネル機能は低下する。(3)糖鎖付加をテュニカマイシンによって阻害すると、細胞膜におけるNaチャネル発現量は減少する。
これらの結果は、Naチャネルの細胞膜発現に様々なシャペロン活性が必須であることを示唆しており、次年度はこれらのメカニズムを詳細に検討する予定である。
また、本研究に関連して、以下の2つの新たな調節機構を解明した。
i)MAPカイネースによるNaチャネルの細胞膜発現調節機構
Extracellular-signal regulated kinase(ERK)の恒常的活性化が、Naチャネルのα-サブユニット遺伝子の安定性を低下させることにより、細胞膜発現を負に調節している。(Mol.Pharmacol.2003.63.in press)
ii)シャペロンによるインスリン受容体の発現調節機構
小胞体におけるインスリン受容体のモノマーからダイマーへの生合成過程には、Hsp90/Grp94活性が必須である。(Mol.Pharmacol.2002.62.847-855)
