海馬神経回路の持続的新生のメカニズムと生理機能の解明

研究概要	大脳辺縁系の一部である海馬体は記憶・学習などの脳高次機能に関与する重要な脳部位である。従来は神経細胞は分裂しないと考えられてきたが、海馬体の神経細胞の約半数を占める顆粒細胞(の前駆細胞)は成体においても増殖することが近年明らかにされている。一方で、顆粒細胞の寿命は数ヶ月程度と短く、成熟後の脳でも顆粒細胞の入れ替わりが継続している。これに伴い、顆粒細胞の作るネットワークもまた短い周期で次々と作り替えられていくと想定される。本研究では、神経細胞としては例外的なネットワーク新生・代替を、「(1)歯状回がなぜ行っているのか」「(2)この特異な現象がどのような機構で生じているのか」「(3)次々に入れ替わってしまう顆粒細胞がどのようにして長期の記憶の保持に寄与できるのか」についての解明を目指している。本年度は特に(2)、(3)において有意義な成果を収めたのでここに報告する。(2)顆粒細胞の生存の維持にアクチン繊維の流動的な再編成の持続が必要であることを示した。また、このアクチン再編成の依存性は、顆粒細胞に特異的であり、かつ、神経活動や細胞外からのカルシウム流入に依存しないこと、また貫通線維からの入力や苔状線維への出力も必要しないことも併せて示した。さらに、苔状線維の軸索の誘導課程に神経伝達物質であるグルタミン酸が必要であるという驚くべき証拠をつかんだ。この現象はグループII代謝型グルタミン酸受容体を介していた。また、苔状線維の形成過程ではまずNMDA受容体のみがシナプス伝達に関与し、引き続いてAMPA受容体も参加するというサイレントシナプスを経由する発達パターンを示すことも突き止めた。(3)苔状線維のシナプス情報は、CA3の連合線維に短期的に、また貫通線維に長期的にtransferされることを明らかにした。前者のケースでは亜鉛イオンが情報転位を媒介していることを示したが、後者の機構は依然として不明であり、さらなる検討が必要である。
