葉緑体から核へ伝達される強光ストレスシグナル伝達経路の解明

研究概要

マイクロアレイを用いた強光に対する転写応答の解析
シロイヌナズナゲノムに含まれている約23,000ある遺伝子のほぼすべてをカバーするGeneChip(22k)を用いて、強光応答遺伝子210個を同定した。また強光ストレスに対する応答のキネティクスやdose-response、そして乾燥ストレス応答、活性酸素に対する応答とのクロストークを解析した。強光特異的に活性化される転写因子・シグナル伝達因子に関しては、それらが特異的な強光ストレス応答を担っている可能性がある。そこで、それらのノックアウト変異体を入手し、強光応答を示すレポーター系統と交配を行い、これらの変異体における強光応答を解析するセットアップを行った。
強光応答に異常を示すシロイヌナズナ変異体の単離と解析
レポーター遺伝子の発現を指標に強光ストレス応答に異常を示すシロイヌナズナ変異体を単離した。これらの変異体からRNAを抽出し、レポーター遺伝子の応答だけでなく内在性の遺伝子発現応答にも異常を示す系統を同定した。また、色素蓄積などの表現型に関しても解析を行い変異体の分類を行った。過剰な強光応答を示す一系統についてファインマッピングを行い、原因遺伝子の同定を試みた。現在のところ同一BAC上に存在する数遺伝子にまで候補を絞ることが出来た。また、さらに炭素イオンビーム処理により変異誘発した系統をスクリーニングし、強光ストレスに対する耐性が低下しかつ強光に対する転写応答異常を示す系統をいくつか同定した。
