造血系転写調節因子GATA-1、PU.1によるマスト細胞分化・成熟機構

研究概要

アレルギー反応の制御を目的にマスト細胞特異的な高親和性IgE受容体(FcεRI)の発現制御機構を解析してきた結果、FcεRIの特異的かつ必須の分子α鎖がGATA-1とPU.1という従来相互に阻害作用を示すと考えられていた転写調節因子により細胞特異的に転写活性化されることを発見した(J.I.168:4546,2002)。このことは基礎的研究の視点から興味深いのみならず、これらの相互作用点が副作用の少ない坑アレルギー剤開発に結びつく可能性を示す。さらにGATA-1の発現量が低下しているGATA-1遺伝子変異マウス由来マスト細胞の解析を行い、FcεRIα、β鎖の転写及び細胞上の受容体発現量が低下することを確認した。一方、PU.1をマウス骨髄由来培養細胞に過剰発現させることにより、マスト細胞と単球系細胞の分岐決定にPU.1発現量が関わっている可能性を発見した(BBRC,313:516,2004;FEBS Lett.561:63,2004)。これらのことから、GATA-1とPU.1はマスト細胞に適当なレベルで共に存在することにより細胞特異的な協調作用を発揮するものと考えられた。加えて、IL-4誘導性のα鎖プロモーターがヒト遺伝子のみ上流に存在していることを発見した(J.I.170:3732,2003)。一方β鎖について、マウスではGATA-1が、ヒトではOct-1とMZF-1が発現に関わる転写調節因子であることを確認してた(J.I.170:334,2003; I.I.15:549,2003; J.I.171:2478,2003)。また、α、β鎖プロモーター中SNPsが転写調節因子の結合能ひいては転写活性に影響を及ぼし、アレルギー疾患発症頻度と有意に相関することを発見し、オーダーメード医療実現に向けて重要な情報と考えている(J.I.171:1927,2003;投稿中)。
