平滑筋細胞におけるムスカリン受容体サブタイプの新たな機能発現モデルの構築

研究概要	本研究では、腸管平滑筋細胞におけるM2およびM3ムスカリン受容体サブタイプ間に存在する相互作用を明らかにし、ムスカリン受容体の新たな機能発現モデルを構築することを目的とした。得られた研究成果の概要は次の通りである。(1)M2サブタイプに連関した非選択的陽イオンチャネル(NSCC)の活性化には、M3サブタイプも同時に刺激されることが必要であるという仮説が考えられている。この仮説を検証するため、各種GTP結合蛋白質(G蛋白質)抗体および情報伝達阻害薬を用いてNSCCに対する影響を検討した。その結果、M3受容体に連関したG蛋白質、イノシトール三リン酸、ジアシルグリセロールなど下流の情報伝達分子よりもむしろ、M3自身から送られる何らかのシグナルがNSCCの活性化に必要であることが明らかになった。この結果を説明するためのモデルとして、M3受容体とM2受容体がヘテロダイマーを形成することによりNSCCの活性化を制御している可能性が考えられた。(2)3種のムスカリン受容体作動薬を用い、各作動薬で生じる収縮反応と収縮反応に対する各種受容体拮抗薬の影響、細胞内Ca^<2+>濃度の変化、収縮蛋白系のCa^<2+>感受性の変化および膜電位変化を比較・解析した結果、これらの作用が上述したM3-M2ヘテロダイマーモデルでよく説明できることを明らかにした。(3)M2またはM3受容体の欠損マウスを用いてNSCCの活性化を検討した結果、M2とM3のどちらの受容体が欠損してもNSCCは活性化されないことが明らかとなった。この結果もM3-M2ヘテロダイマーモデルを支持しており、NSCCには、これまでのモデルとは異なる新しい活性化機構が存在すると考えられる。以上の成果は、2編の学術論文および2編の総説として公表するとともに、第2回キエフ国際平滑筋シンポジウムの招待講演にて発表した。
