細胞間接着による細胞分化の制御機構

研究概要	細胞が極性をもって分化する機構として、ある一つの細胞が、隣接する細胞とのコミュニケーション(細胞間接着)を確立することと、形態形成因子の濃度勾配へ応答することが知られている。この両者の組み合わせによって細胞の分化は制御され、様々な組織・器官が形成されると考えられているが、これらの分子機構は明らかでない。本研究では、細胞分化制御機構の分子機構の解明を目的として、細胞間接着機構ネクチン-アファディン系による細胞分化の制御機構の解析を行ない、本年度は以下の点を明らかにした。まず、ネクチンの阻害物質を作製し、この阻害物質が上皮系細胞のアドヘレンスジャンクションとタイトジャンクションの形成を阻害して細胞極性の形成を障害することを明らかにした。また、ネクチンのトランス結合がRhoファミリーの低分子量G蛋白質であるCdc42とRacの両者を活性化することを明らかにした。さらに、活性化されたCdc42とRacが細胞分化を制御するc-Jun N-terminal Kinaseを活性化することを明らかにした。次に、マウスの精子と卵との受精(接着)部位に、ネクチン-3とアファディンが局在すること、さらに、その部位にネクチン-3とトランス結合するネクチン様蛋白質Necl-5が局在することを明らかにした(投稿準備中)。これらと平行して、アファディンによる外胚葉分化機構の解析のために、Cre-loxPアファディン遺伝子破壊マウスの作製を行なっている。現在、loxP-アファディン遺伝子を持つマウスの作製が終了した。一方、ネクチンによる外胚葉分化機構の解析のためにネクチン-1 ノックアウトマウスを作製した。その結果、ネクチン-1が海馬CA3領域のシナプス形成に必須であることを明らかにした(投稿準備中)。以上の結果から、ネクチン-アファディン系が上皮系、神経系、さらには受精卵の細胞分化と極性形成を制御している可能性が示された。
