研究概要	1)in vivo免疫応答におけるGab2の役割の解析 Gab2 TgにおいてKLH免疫後の抗体産生が抑制されたが、Gads-LATとの結合に必須なPro領域を欠くGab2.DPro Tgマウスでは抑制されなかった。自己免疫疾患モデルである実験的アレルギー性脳脊髄炎(EAE)を誘導したところ、Gab2 Tgとnon Tgとの間には有意な差は認められなかった。Gab2 KOを12 month程度飼育した時点では特に自己免疫疾患様症状は認められていない。これらより、cytokine下流で正のシグナル分子として知られているGab2は末梢T細胞応答において負の制御因子として機能していることが少なくともin vivo抗体産生において示された。 2)TCR Tgとの交配による胸腺の未熟T細胞選択に対する影響の解析 Gab2 Tgマウスはclass I拘束性のpositive selection、negative selectionを抑制するが、Gab2.DPxxxR Tgマウスでは抑制されないことがH-Y TCR Tgマウスとの交配により明らかとなった。また、OVA特異的OT-II TCR Tgマウスにおけるclass II拘束性のpositive selection、peptideによるnegative selectionも同様にGab2 Tgで抑制され、Gab2.dPxxxR Tgマウスでは正常であった。Gab2 KOにおいては、H-Yによるnegative selection、OT-IIにおけるin vitro negative selectionが亢進される傾向を見い出している。これらより、胸腺細胞においても発現が見られるGab2は、胸腺におけるpositive, negative selectionにおいても負の制御因子として機能していることが判明した。
