Decoy DNAによるTh1/Th2サイトカインの発現調節と免疫応答制御の検討

研究概要	免疫応答を制御するサイトカインの産生制御機構の解析は、免疫応答の制御機構のみならず、自己免疫疾患や感染症に対する治療法の開発においても重要な課題である。そこでまず、IL-18刺激によるIL-13産生制御に関わる転写制御のresponsive-elementsの同定を行った。すでにクローニングしたヒトIL-13遺伝子の上流約1kbのDNA断片を用いて、5'-末端から一定領域を欠失させた変異体を作製した。これらのDNAをルシフェラーゼ試験用ベクターに連結し、IL-18刺激でIL-13を産生するマウス胸腺腫細胞、7(3)H細胞、に導入して、IL-18刺激によるルシフェラーゼの発現誘導を検討した。その結果、転写因子の一つであるGATA-3が結合する塩基配列が含まれる領域を欠失させた変異体では、ルシフェラーゼ発現誘導能が失われた。GATA-3を発現しないマウスNK細胞株、LNK細胞はIL-18反応性を示したが、IL-13を産生しなかった。しかしLNK細胞にGATA-3遺伝子を導入すると、IL-18依存性IL-13産生が誘導された。次に、IL-18依存性IL-13産生のシグナル伝達を検討した。7(3)H細胞では、MEKキナーゼ阻害剤の濃度に依存的なIL-13産生の低下が示されたが、p38MAPキナーゼ特異的阻害剤では、それが認められなかった。以上から、IL-18刺激は、p38MAPキナーゼを介してNF-κBへ伝達される従来の経路とは異なる、MEKからp38以外のMAPキナーゼ、例えばSAPKなど、を介してGATA-3に伝達される新たなシグナル伝達経路の存在が明らかとなり、IL-13遺伝子発現を誘導していることが示唆された。しかし、Decoy DNAの細胞内導入を効率良く行なうことができなかった。今後、このDecoy DNA導入に最適な実験条件を検討した上で、サイトカイン産生制御を検討する予定である。
