膵幹細胞分化誘導因子のスクリーニング

研究概要	グルコース応答性インスリン遺伝子転写制御の分子機構に関しては不明な点が多い。細胞内Ca^<2+>の上昇に伴って活性化されるカルモデュリンキナーゼ(CaMK)IVによって転写因子ATF-2のThr^<69>、Thr^<71>、Thr^<73>の3ヵ所のアミノ酸残基がリン酸化される結果、インスリン遺伝子の転写を制御していることを明らかにした(Diabetes 49:1142-1148:2000)。また、転写因子CREMのアイソフォーム(CREM-17X:抑制因子)を単離同定し、糖尿病でその発現が増強している事を明らかにした(J. Biol Chem. 274:21095-21103:1999)。さらに、膵β細胞でTGF-βスーパーファミリー受容体に属するALK-7を単離同定し、これがSmad3を特異的に活性化することにより膵β細胞の発生・分化に関与することを明らかにした(Biochem. Biophys. Res. Commun. 254:707-712:1999)。また、若年性発症の糖尿病であるMODY3の原因遺伝子HNF-1αは転写共役因子p300と直接結合し、糖輸送単体GLUT2の遺伝子発現調節を行っていることを明らかにした(Diabetes 51:1409-1418:2002)。また、消化管ホルモンGIPシグナルの生理的役割を詳細に検討する目的で、GIP受容体欠損マウスを作製した。その結果、このマウスが早期インスリン分泌障害と共に耐糖能異常をきたすことが明らかになった(Proc Natl Acad Sci USA 96:14843-14847:1999)。また、高脂肪食で飼育すると野生型マウスでは肥満を呈しインスリン抵抗性を来たすが、GIP受容体欠損マウスではそのような病態を示さないことから、高脂肪食による肥満やインスリン抵抗性の獲得にGIPが関与している可能性を示した(Nat. Med. 8:738-742:2002)。
