Preconditioningによる肝阻血再灌流障害抑制の検討

研究概要

ヒトおよびラットでNGFI-Bサブファミリーが肝阻血再潅流障害において発現し、かつ障害の程度を反映する遺伝子である事を発見した(Ohkubo T, et al. Early induction of nerve groth factor-induced genes after liver resection-reperfusion injury. Journal of Hepatology 2002)。これらNGFI-Bサブファミリーに属する遺伝子は転写調節因子として働き、アポトーシスに重要な関連を持つとされている。近年肝阻血再潅流障害の抑制法の一つとしてpreconditioning(短時間肝阻血再潅流)が注目されている。しかし、その基礎的背景は今だ明らかでない。preconditioning下における肝阻血再潅流障害抑制効果をNGFI-Bサブファミリー遺伝子をとおして明らかにするのが本研究の目的である。ラット肝阻血再潅流モデルを用いて、その詳細な肝障害機序の解析を行っている。オリゴヌクレオチドプライマーを使いたreverse transcription-polymerase chain reaction(RT-PCR)法にてpreconditioningの有無によるNGFI-Bサブファミリー遺伝子の発現を検討し、internal standard RNAを用いた定量RT-PCR法にて定量している。今後、臨床例でもpreconditioningによる肝阻血再潅流障害抑制効果を検証する予定である。
