ステロイド産生調節因子StARの分解とそのステロイド合成調節における役割-新しいヒト卵巣顆粒膜細胞を用いた研究-

研究概要

ヒト卵巣顆粒膜細胞において、ステロイド産生の律速段階を調節する蛋白Steroidogenic Acute Reguratory Protein (StAR)の分解がステロイド産生をどのように調節するかを検討した。その際、研究に必要なヒトの顆粒膜細胞を十分に確保することが困難であるので、まず不死化細胞株を樹立し研究材料の安定確保を図った。体外受精胚移植の際に得られたヒト顆粒膜細胞に、Ha-rasならびに温度感受性p53を遺伝子導入しステロイド産生能をもつ細胞株を得た(Tajima K et al. Mol Hum Reprod. 2002)。強い卵胞刺激ホルモン(FSH)反応性を有する細胞株HGP53を以後の実験に供した。HGP53に、プロテアソーム阻害剤MG132を投与したところ、細胞内StARの著明な増加と共に、プロゲステロン産生は増加した。この際のStAR増加は、細胞質内およびミトコンドリア内の両方で認められた。このとき、StAR mRNAの発現量に変化はなかった。以上のことから、ヒトの顆粒膜細胞において、プロテアソーム蛋白分解系によるStARの分解がステロイド産生調節に関与していることが示唆された。なお、StARの分解(半減期)は、FSH、フォールスコリンの添加による影響を受けなかった。以上の結果は、顆粒膜細胞のみならずヒト卵巣莢膜細胞株においても確認され、このメカニズムはヒト卵巣のステロイド産生細胞に共通するものと推測された。次に、StARのプロテアソームによる分解がユビキチン依存性であるかを検討するため、抗StAR抗体を用いた免疫沈降後、抗ユビキチン抗体によるユビキチン化の検出を試みた。この結果、StARのユビキチン化を検出することはできず、StARのプロテアソームによる分解は、ユビキチンを介さない経路(アンチザイムなど)により行われる可能性があると考えられた。
