研究概要

本年度は、覚醒動物を用いたin vivo実験系で片側および両側大脳皮質咀嚼・嚥下関連領域の可逆的破壊に伴い摂食・嚥下運動はどのような障害を受けるのかに関するデータ採取を行った。
実験には主にウサギを用いた。実験の第一段階として単一神経活動記録と皮質内微小刺激(ICMS)を組み合わせた手法で脳の機能的マッピングを行うことで、大脳皮質咀嚼・嚥下関連領域の機能的同定を行った。その結果、1)本実験で定義された皮質感覚野顔面口腔領域(Face SI)はH.J.Gould III(J. Comp. Neurol,1986)によって報告されたFace SIの内側部とほぼ一致し、2)顔面運動野(Face MI)および皮質咀嚼野(CMA)の尾側部とGouldの報告によるFace SI外側部は広くオーバーラップし、3)Face MIやCMAとオーバーラップするFace SIへの感覚入力は口腔内あるいは咀嚼筋からのものが多く、体部位局在はやや不明瞭であることが明らかになった。
同定した皮質部位を冷却法を用いて可逆的に破壊したところ、以下のことが明らかになった。
(1)Face MI(CMAとのオーバーラップ領域を含む)の片側性破壊により、動物の食物咀嚼側は、破壊以前の咀嚼側に関わらず、皮質破壊側と同側(機能残存皮質の支配側)となる。
(2)Face MIの両側性破壊により、摂食行動の開始が著しく障害され、咀嚼運動の遂行が不可能となる。
以上のことはFace MIは摂食行動の開始に関して重要な脳部位であるとともに、咀嚼側の決定に関与していることを示唆している。片側のFace MI破壊は摂食行動の開始そのものには影響を及ぼさなかったことから、摂食行動の開始に関する機能は対側のFace MIによって速やかに代償されうるが、咀嚼側の決定に関する機能は代償されないことが示唆された。
