研究概要

ヒト生体資料を対象とした外因性内分泌活性化学物質の理化学的分析手法を構築するため、液体クロマトグラフ/質量分析法(LC-MS)やガスクロマトグラフ/質量分析法(GC-MS)を用いて、検討を行った。対象とする生体資料は、尿や血液とし、その前処理に関して、カラムスイッチング法やTwister加熱脱着法などを応用した。また、外目性内分泌活性物質の評価として、ヒト乳がん細胞や核内受容体結合試験を利用して、対象を絞った結果、アルキルフェノール類(ノニルフェノール、オクチルフェノール及びビスフェノールA)や可塑剤(フタル酸ジー2-エチルヘキシル)などを候補として挙げることができた。本化合物は、様々なプラスチック製生活用品に利用されているため、工業的にもその生産量は多く、職業上暴露される可能性が十分高いことが示唆される。また、来年度に関しては、対象化合物を増やし、一斉分析を可能とすることを目指す。
本年度においては、ヒトの尿・血液を利用して、アルキルフェノール類と可塑剤の暴露モニタリング法を構築した。多量の検体数を短時間で分析するため、煩雑な前処理などを省略できるカラムスイッチングLC-MS法を活用した。本法の詳細な分析条件を検討した結果、pptレベルでの高感度分析が達成でき、十分に化学物質暴露モニタリングに応用することが可能と思われた。それに加え、代謝物についても考慮した結果、抱合体解離酵素(グルクロニダーゼなど)の最適条件を検討し、本測定とモニタリングが十分に可能となった。
