生体膜透過性ペプチド誘導体を配位子とするインスリン様バナジウム錯体の設計と評価

研究概要	今年度は、(1)ピコリン酸もしくはマルトール誘導体をリード配位子とした新しいバナジル錯体の合成、続いて(2)バナジル錯体のインスリン様作用の評価と構造活性相関性、最後に(3)in vivoにおけるラット消化管吸収評価法の開発を中心に検討した。第一段階として、N_2O_2もしくはO_4配位形式の錯体を合成するため、ピコリン酸もしくはマルトールをリード化合物とし、疎水性増大効果にもとづいて細胞内取り込みを促進させ、かつ、小腸粘膜からの吸収を上昇させるようにアリル基を導入した新規配位子を合成し、続いて、バナジルイオンと反応させて錯体を合成した。得られた12種類の錯体の物理化学的特性を評価するとともに、化学構造を推定した。第二段階として、新たに合成された錯体について、エピネフリン刺激によるラット脂肪細胞からの遊離脂肪酸放出に対する抑制効果を指標としたin vitroにおけるインスリン様活性を評価し、IC_<50>値を指標として構造活性相関性について考察した。その結果、各錯体のインスリン様活性を決定する重要な物性パラメータはLogP値であると分かった。ここで、各錯体のLogP値は水/クロロホルム系から評価した数値を用いた。以上より、中間的な同じ安定度定数(N_2O_2もしくはO_4配位型)を有した中性錯体の場合、受動拡散を介した膜透過によって細胞内へ取り込まれるため、アリル基導入による錯体の疎水性増大(LogPの上昇)がインスリン様活性の上昇に結びつくと分かった。第三段階として、錯体の消化管粘膜透過性については、培養細胞もしくはin situラット消化管を用いた方法では正確に評価できないことが分かったため、in vivoにおける直接的な消化管吸収解析法を開発した。これは、門脈血と循環血を同時に採取して両サンプル間の薬物濃度差から吸収性を評価する新規動態解析法である。本方法を用いて、各錯体の消化管吸収特性の検討は、現在継続中である。最終的には、粘膜透過性に対する疎水性基導入による促進効果の定量的評価にもとづいて、Bioavailabilityを指標とした構造-消化管吸収相関性について考察し、経口投与により有効性を発揮するための最適な錯体構造を決定する予定である。
