研究概要

ホスホエノールピルビン酸カルボキシラーゼ(PEPC)は、C4植物で光合成初期炭酸固定反応を触媒している酵素である。PEPCをイネに高発現させることによって、炭酸固定効率が増大し、光合成の酸素阻害が低減されるとの報告があったが(Nature Biotech.17,76-80(1999))、その後の報告で、イネで高発現したPEPCは必ずしもその酵素活性を最大限に発揮していない可能性が示唆されている。PEPCの活性調節がうまく機能していないのが原因であると考えられている。そこで、我々はPEPCの活性調節機構の全容解明に向けてPEPC-PKの結晶構造解析を目指ざし、以下の実験を進めた。
C4植物フラベリア由来PEPC-PKの大量精製系を二種類(pET16b,His-tagged酵素とpeT43a,thrombin処理酵素)確立し、大量精製に成功した。その後の恒温インキュベータ(20℃と4℃)で、結晶化を行ったところ、0.05mm程度の大きさの結晶を作成することに成功したが、SPring-8にてX線回折実験を行ったところ回折能8.0Å程度と構造解析には不十分なデータしか得られなかった。更なる結晶の品質向上を目指し、溶液状態を制御による結晶の品質およびサイズを向上させる技術を開発した(Jpn.J.Appl.Phys 43,L79-L81(2004))。現在、本技術を用いて結晶化を行っている。
更に、PEPCの立体構造に基づくアロステリック活性調節機構に関する論文1報と総説2報を報告した(Structure 10,1721-1730(2002),Arch.Biochem.Biophys.414,170-179(2003),Annual Review Plant Biol.(2004)in press.)。
また、PEPCはリンゴ酸などの有機酸によって活性阻害されることが知られているが、生体内でそのリンゴ酸を生成する酵素NADP依存型リンゴ酸酵素の精製・結晶化にも取り組んだ。NADP-リンゴ酸酵素については、微結晶を得ており(0.1mm程度)、5.0Å分解能の回折能を示す結晶を得ている。
