研究実績の概要 |
The first year of this project aims at fabricating 3D co-culture artery-on-a-chip to mimic the vascular microenvironment in the healthy and disease states. Two simple and robust fabrication methods were successfully developed and compared: (i) in-channel bilayers of smooth muscle cells (SMCs) and endothelial cells (ECs), and (ii) separated multilayers of bulk-SMCs with in-channel monolayer of ECs. Optical and fluorescence microscopy imaging illustrated the cell sprouting behaviours of both cells within the bulk matrix and along the contour of the microchannels, which resembled the in vivo functions. Continuous efforts are underway to incorporate various pathological factors (pro-inflammatory TNF-α, LDL-cholesterols and monocytes) to induce vascular inflammation and foam cells formation. Successful illustration of the diseased models is anticipated within the next 2-3 months and will lead to further conference presentations and an article publication.

今後の研究の推進方策 |
Within the next 2-3 months, successful illustration and comparison of the healthy and diseased vascular models is anticipated to lead to further conference presentations and article publication in a peer-reviewed journal. In the next 10-12 months of study, the functional vascular disease-mimicking tissues will be utilized to investigate the anti-atherosclerotic therapeutic effects of several pharmaceutical compounds (statin drugs, cyclodextrins).
