消化器癌の転移とその治療の研究

研究概要	本研究課題においては、消化器癌の転移、特に周辺リンパ節への転移の分子機序を明らかとし、これに関わる遺伝子の変異モデルマウスを作製し、発癌から転移までの過程を再現するモデルマウスを構築する。これを用いた解析により、化学療法による転移抑制の標的分子を同定し、新しい科学療法薬開発のための戦略を確立することを目的とする。今年度は、可移植性癌細胞を用いたマウス転移モデルにより、転移に関与する新たな因子の同定と、その作用機序の解析を中心に行なった。最近、ケモカインが走化性作用により癌転移に関与しており、特にCXCR4受容体を介したシグナルが転移に重要であることが報告された。我々も複数の癌細胞株を用いて、全ケモカイン受容体の発現プロファイルをmRNA定量により解析した結果、CXCR3の発現上昇が認められた。さらにCXCR3リガンドのIP-10あるいはMIGに対する中和抗体は、B16F10メラノーマ細胞のリンパ節転移を抑制したので、CXCR3を介したシグナルが転移に重要であることが明らかとなった。一方、リンパ節転移には新たなリンパ管新生が必要であるが、分子機序は明らかにされていない。マウス生体組織を用いて、リンパ管新生過程を経時的・形態的に解析するためにLYVE-1に対する抗体を作製した。得られた抗体を用いた免疫染色の結果、リンパ管特異的に反応したので、今後CXCR3シグナルとリンパ管新生との関係についての解析に使用する。また、癌転移にプロスタグランジン合成酵素であるCOX-2が関与している可能性が報告されている。そこで、ケモカイン産生を含めた炎症反応と転移の関係を解析する目的で、COX-2およびPGE_2合成酵素であるmPGESのトランスジェニックマウスを作製した。更に、Peutz-Jeghers氏病の原因遺伝子LKB1のマウスホモログをノックアウトしたところ、Peutz-Jeghers氏病に特有な胃や十二指腸のハマルトーマを形成したのみならず、肝臓に肝細胞癌(HCC)を発生し、その一部は肺に自然転移を起こすことを見出した。今後、これらの転移モデル、抗体、マウスモデルを用いて、転移過程に関与する分子機序を明らかにして行く。
