消化器癌の転移とその治療の研究

研究概要

本研究課題においては、消化器癌転移の分子機序を明らかにし、これに関わる遺伝子の変異マウスを作出して発癌から転移までの過程を再現するモデルマウス実験系を構築する。この系を用いて転移抑制の標的分子を同定し、新しい化学療法薬開発のための戦略を確立することを目的とする。前年度にB16F10メラノーマ細胞のリンパ節転移がケモカイン受容体CXCR3の中和抗体で抑制されることを明らかにしたが、今年度はさらに解析を進め、CXCR3アンチセンス発現細胞を用いてCXCR3のリンパ節転移における重要性を遺伝学的に証明した。また、この系では完全フロイントアジュバントが転移を促進することから、癌の非特異的免疫療法には危険が伴う可能性が示唆された。CXCR3が多くの大腸癌細胞株で発現されていることも見出したので、今後CXCR3の大腸癌転移への関与を検討する。さらにColon26大腸癌細胞のマウス転移モデルを用いて、リンパ節及び肝臓の転移巣の細胞を再び移植することを繰り返して高転移細胞株を樹立した。現在、DNAマイクロアレイ及びプロテオミクスの手法で高転移株と親株とを比較し、転移に関わる分子の同定を進めている。一方、家族性大腸腺腫症のマウスモデルのApcΔ716マウスにホメオボックス転写因子Cdx2のヘテロ接合変異を導入したApcΔ716Cdx2+/-複合変異マウスを作出したところ、大腸にポリープを多発してよりヒトの疾患に近い形質を示すことを見出した。また、その機序として、Cdx2の低下がmTOR経路活性化を介して染色体の不安定性を亢進させ、LOH(loss of heterozygosity)の頻度が上昇することを示した。Cdx2の発現は大腸癌悪性化に伴い低下することが報告されており、今後Cdx2の発現低下、mTOR経路活性化による染色体異常が癌細胞の転移能獲得に果たす役割について解析を加えていく。
